AN IMPROVED METHOD OF EXTRACTING GENOMIC DNA FROM PRESERVED TISSUES OF JASMINUM SPP. FOR AFLP AND SSR STUDY

In this study, an improved method of genomic DNA extraction from six months old dried leaf tissue of different Jasminum spp in India is described. Two widely used DNA extraction protocols Dellaporta (1983) and Doyle and Doyle (1990) were compared across 56 cultivars and wild types of Jasminum. The modified Doyle and Doyle protocol yielded high quality and quantity of DNA, i.e. average of 1200 to 1500 ng/g of leaf tissue. RNA and protein contamination was minimal with this method. Results from 56 plants were examined. The DNA so extracted was found useful for different marker analysis like AFLP and SSR and consistently amplified all across 56 plants from same and different species of Jasminum. The protocol yielded similar quality and quantity of DNA as extracted from fresh tissues.
